Detection methods range from physical (e.g., nanogold) to enzymatic (horseradish peroxidas, alkaline phosphatase) to fluorescent (the traditional FITC or newer Quantum Dots) and are selected based on the type of microscopy that you will be performing and the sensitivity required for your imaging. Each method has its advantages and disadvantages, and the use of one over another is often as much a matter of preference and artistry as strictly scientific.
